Though the nuclear transfer methods have been known for 40 years and have been widely discussed in reproductive medicine since 2017, there are a lot of unpublished technical nuances that a worker is doomed to face in case they decide to implement this technology in clinical or research practice. Having started in 2019, we encountered many practical difficulties that had to be overcome with long and painstaking work. In total, we have so far performed more than 1,000 pronuclear transfers and obtained blastocysts, successful in vitro implantation, as well as pregnancies and live births in mice. Our scientific interest is investigating the heteroplasmy dynamics and distribution among tissues after nuclear transfer. However, this report will focus mainly on methodological tips and tricks.
