Analysis of any single PBA sample with multiplex microspheres and corresponding detecting antibodies in comparison with the blank, a significant bead counts decrease was revealed not only within the corresponding PBA monoplex, but also in most of the other monoplexes. Thus 80% of the comparisons this decrease was statistically significant (Fig. 1). Less often this effect observed in the presence of RUB, and the most pronounced difference was noted in the presence of TUL. The greatest bead counts decrease counts for STM (-79%, from 528 to 109), PAE (-64%, from 570 to 250) and SHA (-92%, from 560 to 48) was observed during detection of self-antigens. The most pronounced decrease to the bead counts for all monoplexes induced by gram-negative bacteria (from -20% to -50%). Fully multiplexed immunoassay in the presence of 9 PBA (PBS-TBN as assay/wash buffer) showed significant bead count decrease for all nine regions compared to the blank: for PAE - by 40%, for ADV - by 48%, for TUL - by 49%, for RUB - by 51%, for SAU by 60%, for CTB by 67%, for STM by 79%, for BRU by 85% and for SHA by 91%. Change of the buffer to TBN+ (specify changes) lead to significant improvements for most of the targets PAE - by 45%, for ADV - by 36%, for TUL - by 23%, for RUB - by 39%, for SAU by 37%, for CTB by 39%, for STM by 55%, for BRU by 35% and for SHA by 64%.
