Phosphate-buffered saline (PBS) without Ca+2 and Mg+2 is a buffer solution commonly used in biological research. PBS has many uses because it is isotonic and non-toxic to most cells. It is used often for rinsing cells from culture medium and different reagents and to preparing living cell samples for different types of analysis. But it was found what PBS is not suitable for all cells. Exposure of the cultured pluripotent human embryonic stem cells (hESCs) to Ca+2 – Mg+2 – free PBS for 20 min resulted in extensive dissociation, blebbing and cell death.
