В настоящее время во всем мире ведется активная разработка методов получения наночастиц для переноски различных веществ, в первую очередь, для контролируемой доставки лекарственных препаратов, а также для создания нанореакторов, искусственных органелл и других приложений [1]. Одним из способов инкапсуляции веществ является их помещение внутрь везикулоподобной капсулы. Везикулы могут спонтанно формироваться в растворе за счет гидрофобных взаимодействий: так формируются липосомы на основе липидов [2, 3] и липидо-подобных молекул [4], имеющих гидрофильную и гидрофобную части. Увеличить стабильность везикулярной структуры и получить возможность управлять ее размером и толщиной оболочки позволяет использование вместо липидов молекул блок-сополимеров, имеющих гидрофильный и гидрофобный блоки [5]. Другим фактором, способствующим самоорганизации молекул в растворах, являются электростатические взаимодействия. Например, полые нанокапсулы можно получить путем последовательного осаждения слоев полиэлектролита на заряженную коллоидную подложку и последующего растворения этой подложки [6]. Везикулы конструируют из блок-сополимеров, имеющих нейтральный и заряженный блоки (PICsomes [7]); с целью улучшения контроля свойств таких везикул в их оболочке проводят реакцию сшивания [8]. Также, для создания наноконтейнеров разрабатывают чувствительные к внешним воздействиям полиэлектролитные микрогели со структурой рыхлое ядро–плотная оболочка, образуемой за счет электростатических взаимодействий [9], вызывающих концентрирование заряженных мономерных звеньев в приповерхностном слое. Данные экспериментов показывают возможность получения водорастворимых стехиометрических интерполимерных полиэлектролитных комплексов: в частности, с использованием гомополимеров, содержащих гидрофильные группы [10]. Теоретический анализ позволил предположить, что растворимость таких комплексов обусловлена формированием структуры ядро – оболочка, в которой вокруг ядра образуется гидрофильная корона [11]. Предварительные оценки показали, такие комплексы способны формировать более сложные структуры, такие как полые везикулоподобные нанокапсулы сферической и продолговатой формы, если макроионы обладают существенно различным сродством к растворителю, отличаются плотностью распределения заряда, а также когда их суммарные заряды нравны (нестехиометрические комплексы). Для везикулоподобных структур наиболее простым способом получения и эффективного управления их размером и размером полости внутри них было бы выгодно использовать минимальное количество макромолекул. Так, в амфифильных на уровне отдельного звена гомополимерах каждое повторяющееся звено содержит гидрофильные и гидрофобные группы. Таким образом, небольшие амфифильные фрагменты соединены химическими связями в единую цепь, и достичь их высокой концентрации можно путем ухудшения качества растворителя при малой концентрации самого полимера в растворе. Методами компьютерного моделирования и аналитической теории ранее нами было показано, что макромолекулы, амфифильные на уровне отдельного звена, в плохом растворителе способны образовывать различные микроупорядоченные структуры, в том числе и везикулы, которые стабильны в широком интервале исследованных параметров макромолекулы и качества растворителя [12]. Эти результаты позволяют предположить, что образование полой везикулярной структуры может происходить также и в полиэлектролитном комплексе, состоящем из небольшого числа макромолекул с различным сродством к растворителю. В рамках данного проекта будет проведено компьютерное моделирование и теоретическое исследование структуры полиэлектролитных комплексов противоположно заряженных макромолекул, обладающих разным сродством к растворителю, в частности, будут определены условия формирования везикулоподобных структур и структур типа ядро-оболочка. Компьютерное моделирование и теоретический подход позволят исследовать детали строения полиэлектролитного комплекса, недоступные экспериментальным методам. 1. L. K. Muller, K. Today, active development of methods for obtaining nanoparticles for the transport of various substances is being carried out worldwide, primarily for the controlled delivery of drugs, as well as for the creation of nanoreactors, artificial organelles and other applications [1]. One way of encapsulating substances is to place them inside a vesicle-like capsule. Vesicles can be spontaneously formed in solution due to hydrophobic interactions: thus liposomes based on lipids [2, 3] and lipid-like molecules [4], which have hydrophilic and hydrophobic parts, are formed. To increase the stability of the vesicular structure and to be able to control its size and shell thickness, one can use block copolymers with hydrophilic and hydrophobic blocks instead of lipids [5]. Another factor contributing to self-organization of molecules in solutions is presence of electrostatic interactions. For example, to produce hollow nanocapsules a method of sequentially depositing polyelectrolyte layers on a charged colloidal substrate and then dissolving this substrate is used [6]. Vesicles are constructed from block copolymers having a neutral and charged blocks (PICsomes [7]); in order to improve the control of the properties of such vesicles, a crosslinking reaction is carried out in their shell [8]. Also, to create nanocontainers, sensitive polyelectrolyte microgels are developed with the structure of a loose core-dense shell, formed due to electrostatic interactions [9], causing concentration of charged monomer units in the near-surface layer. The experimental data show the possibility of obtaining water-soluble stoichiometric interpolymer polyelectrolyte complexes: in particular, using homopolymers containing hydrophilic groups [10]. Theoretical analysis suggested that the solubility of such complexes is due to the formation of a core-shell structure in which a hydrophilic corona is formed around the core [11]. Preliminary estimates demonstrated, that such complexes can form more sophisticated structures, such as hollow vesicle-like nanocapsules of spherical and oblong shape, if macroions have substantially different affinity for solvent, different density of charge distribution, and also when their total charges are unequal (non-stoichiometric complexes). For vesicle-like structures, the most advantageous way to obtain them and effectively control their size and cavity size inside them would be to use the minimal amount of macromolecules. Thus, in homopolymer amphiphilic on a level of an individual repeated unit, each unit contains hydrophilic and hydrophobic groups. So, small amphiphilic fragments are connected by chemical bonds into a single chain, and their high concentration can be achieved by changing solvent quality at low concentration of the polymer itself in the solution. By means of computer simulation and analytical theory, we have previously shown that macromolecules, amphiphilic at the level of an individual repeat unit, in poor solvent are able to form various micro-ordered structures, including vesicles, which are stable in a wide range of the investigated parameters of the macromolecule and solvent quality [12]. These results suggest that the formation of a hollow vesicular structure can also occur in a polyelectrolyte complex consisting of a small number of macromolecules with different affinity for the solvent. Within the framework of this project, computer simulation and theoretical research of the structure of polyelectrolyte complexes of two oppositely charged macromolecules with different affinity for the solvent will be carried out, in particular, the conditions for the formation of vesicular-like structures and core-shell structures will be determined. Computer modeling and theoretical approach will allow us to investigate the details of the structure of the polyelectrolyte complex, which are inaccessible to experimental methods. 1. L. K.
