Целью настоящего Проекта является создание научной платформы для разработки медицинских тераностических наноагентов с высокой эффективностью и биосовместимостью. Для достижения указанной цели необходимо решить следующие конкретные задачи: 1. Получение с помощью спектроскопии когерентного антистоксового рассеяния света (КАРС) принципиально новой информации о взаимодействиях наночастиц с окружающими биомолекулами, которая обеспечит решение следующих конкретных задач: - изучение влияния взаимодействий поверхностных групп наночастиц/корон нанокомплексов на состояние и функциональную активность прикрепленных и взаимодействующих с поверхностью наночастиц биомолекул; - изучение влияния взаимодействий поверхностных групп наночастиц/корон нанокомплексов с окружающими биомолекулами на свойства наночастиц-носителей (в первую очередь, на фотолюминесценцию). 2. Разработка метода контроля выведения наночастиц из организма по спектрам их фотолюминесценции. Успешное решение указанных задач Проекта обеспечит научную основу для разработки тераностических наноагентов с высокой эффективностью и безопасностью для живых организмов. Речь идет о многофункциональных наноагентах, одновременно выполняющих функции фотолюминесцентного маркера и носителя лекарственных препаратов. Именно такие тераностические наночастицы/нанокомплексы способны ускорить переход к персонализированной медицине, которая заключается в персонализации и оптимизации профилактических мероприятий и лечения пациентов для исключения негативных последствий и осложнений, проявляющихся из-за индивидуальных особенностей. Создание наночастиц, обладающих стабильной фотолюминесценцией, которая не тушится в результате взаимодействия окружающих биомолекул с поверхностью наночастиц, а также одновременно надежно удерживающих на своей поверхности лекарственные препараты и осуществляющих их адресную доставку, препятствуя попаданию препарата в здоровые ткани, открывает широкие перспективы для эффективного лечения многих заболеваний, в первую очередь, онкологических. При этом решается проблема строгого дозирования лекарственных препаратов и их распределения для исключения негативных побочных эффектов в биоткани. Кроме того, сама концепция применения тераностических наноагентов подразумевает возможность контроля распределения лекарств в организме, движения наночастиц и их выведения. Поэтому одной из основных задач настоящего Проекта является развитие чувствительного метода оптической визуализации тераностических наноагентов в биологических тканях и мониторинга выведения наночастиц из организма. Успешное решение этой задачи также обеспечит высокую степень безопасности предлагаемых медицинских наноагентов для живых организмов. Таким образом, тематика Проекта полностью соответствует указанному направлению из Стратегии НТР РФ.
The relevance of the research planned in the Project is directly related to the wide range of applications of nanoparticles in biology and medicine: creation of highly effective luminescent biomarkers, development of novel nanotechnologies and nanomaterials for targeted drug delivery, study of the transport of nanoagents and biomacromolecules, study of biocompatibility and influence of nanoagents on the properties of complex biological systems, including animal and human organisms. At present, significant progress has been made in this direction: methods for the synthesis of complexes based on nanoparticles of different nature (carbon, silicon, etc.) that can perform the required functions have been developed, and many nanoparticles and nanocomposites have been successfully tested on cell lines and animals. However, for widespread use of novel theranostic nanoagents one needs to be sure of their high biocompatibility, i.e. understand what changes at the molecular level occur with biomolecules in the body as a result of their interaction with the surface of nanoparticles or their crowns (coatings). In addition, these interactions can have different effects on the properties of the nanoparticles themselves, for example, to enhance and extinguish their photoluminescence. Obviously, to solve these problems, it is necessary to thoroughly study the mechanisms of processes that occur when the surface of nanoparticles and nanocomplexes (including their protein crowns or attached drugs) interacts with surrounding molecules in the biological environment (first of all, these are water molecules and biomolecules – proteins, DNA, lipids, etc.). The project is aimed at creating a scientific platform for the development of medical teranostic nanoagents with high efficiency and biocompatibility. This implies obtaining fundamentally new information about the mechanisms of interactions of nanoparticles with surrounding biomacromolecules and the effect of these interactions on the properties of nanoparticles and the biological environment using coherent anti-Stokes Raman scattering (CARS) spectroscopy. To successfully complete the Project, it is necessary to solve the following specific tasks: 1. Obtaining fundamentally novel information about the interactions of nanoparticles with surrounding biomolecules using CARS spectroscopy. 1.1. Study of the mechanisms of processes occurring during the interaction of the surface of nanoparticles and nanocomplexes (including their protein crowns or attached drugs) with environmental molecules in the biological environment (first of all, these are water molecules and biomolecules – proteins, DNA, lipids, etc.). 1.2. Study of the effect of the described interactions on the physiological state and functional activity of biomolecules and drugs, which is necessary for the development of effective and safe medical nanoagents. 1.3. Investigation of the effect of interactions of nanoparticles’ and nanocomplexes’ surface with surrounding molecules in aqueous suspensions and in the biological environment on the properties of nanoparticles (including photoluminescent, colloidal, and adsorption ones). 2. Development of a method for optical visualization of nanoparticles and determination of their concentration in biological tissue to control the removal of nanoparticles from the body (for example, with urine). It is assumed that CARS will provide a successful solution to this problem due to its high selectivity to certain vibrational transitions and high sensitivity to small concentrations of impurities in the substance. In the framework of the project it is for the first time proposed to implement such a sophisticated, but still highly sensitive method of CARS spectroscopy to study the effect of nanoparticles injected into the biotissue on the state and functionality of nanoparticles, surrounding and interacting with surface groups or their biomolecular crowns (coatings of nanocomplexes). It is supposed to solve the problem of monitoring the removal of nanoparticles from the body with urine at a higher level. As described in section 4.5, the CARS method allows to obtain fundamentally new information about the interactions between even single molecules, and high selectivity and sensitivity of the method makes it possible to determine the concentration of impurities in a substance with an accuracy several orders of magnitude higher than spontaneous Raman spectroscopy allows. The project executors’ experience and background in the field of vibrational spectroscopy of aqueous media, dispersed systems, nanoparticles and biological systems, as well as experience in solving inverse problems of optical spectroscopy using modern machine learning methods (see section 4.7) gives confidence in the successful solution of the tasks set in the project and the possibility of obtaining the planned results.
