Аннотация:Conditions of the endo-1,4-.beta.-glucanase biosynthesis in an extremely thermophilic strictly anaerobic non-sporulating bacterium, strain Z 1320, were studied. The highest level of endoglucanase activity was detected, when the bacterium was cultivated at 70-75.degree. C, pH 7.0 on a medium with insoluble substrates-microcrystalline cellulose and filter paper; a lower level was observed in case of cellobiose, glucose and glucose 6-phosphate. Organic additive (vitamins, yeasts and maize extracts) repressed the endoglucanase biosynthesis. On 0.1-1.0% cellulose as a sole organic carbon source, the maximal rate of endoglucanase formation was observed up to 4 days of cultivation and coincided with the exponential phase of biomass accumulation. To this moment, from 80 to 19% of the substrate was solubilized (0.8-1.9 g/L); the final endoglucanase level being 0.017-0.013 U/ml, respectively, 3.3 and 40% of endoglucanase was adsorbed on 0.1% and 1% cellulose of the growth medium, respectively, and less than 1% of the total enzyme was located on the cell surface and inside the cells.
