Аннотация:The functional expression of the Na/K pump(measuresd as ouabain-sensitive Rb influx), 3[H]ouabaun binding as well as the expression of alpha-1-subunit of Na/K ATPase pump was determined in HBLs activated by phytohemagglutinin (PHA), phorbol ester (PDBu) and ionomycin (I) or PDBu and interleukin-2 (IL-2). The number of ouabain-binding sites was found to increase five fold in actiavted HBLs as compared with the resting HBLs. The ouabain-sensitive Rb influx per cell increased in time-dependent manner being maximal (ten fold increase) at 48 h of mitogen stimulation. There was a strict correlation between the long-term enhancement of ion pump and the ouabain-binding level during the late G0/G1/S progression. The immunosuppressive drug cyclosporin A inhibited the Il-2-dependent up-regulation of Na/K pump. The results suggest that in mitogen-induced HBLs the functional expression of the Na/K pump is closely associated with the cell cycle progression; the cell-cycle-associated enhancement of ion pump is a result of increased number of newly synthesized functioning Na/K pumps in cell memebrane.
